Las oraciones en presente simple afirmativo solo se cambian cuando el sujeto es She, He o It. Se le agrega una s, es o ies depende del verbo, por lo que cuando se pregunta o es una oracion negativa, se usa el auxiliar does
Ejemplo:
She/He works in a hospital
She doesn't work in a hospital
Does she work in a hospital?

Fijate que en el negativo y en el interrogativo se usa DOES pero al verbo se le quita la S, ES o IES, eso es importantisimo.
DOESNT en el negativo por que significa DOES NOT.

Cuando el sujeto es I, We, They, You, no se le agrega nada en la oracion afirmativa, pero se utiliza el auxiliar DO cuando es negativo o pregunta.
Ejemplo:
I work in a hospital
I dont work in a hospital
Do I work in a hospital?
Al verbo no se le agrega nada. DONT en el negativo porque significa DO NOT
